I think from all the stuff I've read online, the letter has been asked for once or twice, but not asked for the other hundreds of thousands of times people have requested a business visa. I've had a letter (real) both times I entered Cambodia for work without an existing visa. The first time I crossed at Bavet on the sinh tourist bus and they didn't want to to help get the e type visa - everyone else on the bus was getting a tourist visa - so I had to go to the immigration booth and show them the letter. I suspect I could have still got it without the letter as the guy didn't actually read it, barely glanced at it and said ok. The second time was with a new passport at pochentong, no letter required.
