For Phnom Penh to Kampot, consider getting a taxi. Last time we went, I think it cost $35 to get there. If you've got accommodation booked, the cab will take you to the door. Big bonus, no smelly backpackers to deal with, and the taxi drivers we've used drive more sanely than the bus drivers.
Conversely, if you went on say, Giant Ibis bus co, IIRC the tickets are about $10 each way, you get dumped near the durian roundabout and have to make your own way to your hotel. There would be at least one stop on the way which adds time to the journey, whereas with the cab, he'll probably stop for fuel once and that should be it.
