Can I send money from an Australian bank in AU dollars and get it from Acleda bank in US dolars and would the exchange rate be better doing it this way rather than getting it exchanged first in Australia?

You need to study the charges and exchange rates applied by your bank in Australia.

Have a look at Transfermate. In both Ireland and the UK these guys work out much cheaper than the banks. You need to be moving over a grand each time to get the best rates though.

I've never transfered money from Aus to Cambo so really can't comment. But I've sent plenty of money from Aus to Thailand. I've always sent Aus $ to my Thai bank account who then converted them to Baht, much better rates that way.
I doubt that this scenario would be any different in Cambodia.

The exchange rate with ANZ Aus is pretty shit though usually, also the Aus bank will charge an international transfer fee, and Acleda charges a receiving fee. Best to send large sums rather than anything less than $USD1000.

Ozforex, only $15 a transfer, one gets the rate and they have extremely helpful telephone staff.
